Abstract
Methodologies for analysis of proteins isolated from perennial tissues are described, with particular reference to overcoming problems related to protein degradation and presence o phenolic compounds. Techniques described cover the basic steps of protein isolation, quantification and characterisation using one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Also described is a technique which permits high-resolution protein characterisation, two-dimensional iso-electric focusing (IEF) electrophoresis, where protein is separated in the first dimension by IEF according to charge (iso-electric point, pl), then in the second dimension by SDS-PAGE, where protein is separated according to size (molecular weight, MW). In addition, two immuno-detection techniques, Western blotting and immuno-localisation are described in detail.
